Specificity This kit recognizes natural and recombinant Chicken HPX. No signif icant cross-reactivity or interference between Chicken HPX and analogues was observed. Note: Limited by existing techniques, cross reaction may still exist, as it is impossible for us to complete the cross-reactivity detection between Chicken HPX and all the analogues.

Protocol This ELISA Kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with HPX. During the reaction, HPX in the sample or standard competes with a fixed amount of HPX on the solid phase supporter for sites on the Biotinylated Detection Ab specific to HPX. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of HPX in the samples is then determined by comparing the OD of the samples to the standard curve. 6th Edition, revised in June, 2015 www.elabscience.com 4

Storage Comment All the reagents in the kit should be stored according to the labels on vials. Unused wells should be returned to the foil pouch with the desiccant pack and resealed along entire edge of zip-seal. Substrate Reagent shouldn't be kept at -20°C (Check!). Exposure of reagents to strong light should be avoided in the process of incubation and storage. All the taps of reagents should be tightened to prevent evaporation and microbial contamination.
