Specificity
|
The reactivity of the antiserum is directed to the major isotypes of the rat immunoglobulin system (classes and both light chain types) including antibodies to common determinants, to class and to the surface determinants of the common Fab portion, as tested by immunoelectrophoresis and double radial immunodiffusion against pooled serum and purified immunoglobulins. In immunoelectrophoresis and double radial immunodiffusion using various antiserum concentrations against normal rat plasma and serum, the characteristic IgG, IgA and IgM precipitin lines are obtained.

Tetramethylrhodamine isothiocyanate-conjugated IgG fraction of polyclonal goat antiserum to rat IgG, IgA and IgM, heavy and light chains, adsorbed for human and mouse cross-reactivity. Fluorochrome/IgG protein molar ratio (F/P) is approximately 1.7.

Purification
|
Immunoaffinity adsorbed using insolubilized Ig-depleted rat serum as required, to eliminate antibodies reacting with other serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum. This product has been passed over appropriate immunoadsorbent to remove antibodies cross-reacting with human and mouse immunoglobulins. This renders it specific for use in test systems containing material of human and mouse origin. Hyperimmune antisera with strong precipitating activity are selected for fractionation and purification of the IgG (7S) fraction containing the bulk of the defined antibody specificity. It is free of other serum proteins as tested by immunoelectrophoresis.
