Immunogen Highly purified IgG, including all subclasses, and pools of homogenous IgA and IgM isolated from mouse serum. Freund's complete adjuvant is used in the first step of the immunization procedure.
Isotype IgG
Specificity The defined antibody specificity is directed to the major isotypes of the mouse immunoglobulin system (classes, subclasses and L chain types) including antibody to common determinants, to class and subclass-specific determinants, and to the surface determinants of the common Fab portion as tested against as tested in immunoelectrophoresis and double radial immunodiffusion (Ouchterlony).
Characteristics Purified IgG fraction of polyclonal goat antiserum to mouse immunoglobulins IgG, IgA, IgM, heavy and light chains, adsorbed for human cross-reactivity.
Purification Immunoaffinity adsorbed using insolubilized Ig-depleted mouse serum as required, to eliminate antibodies reacting with other serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum. This product has been passed over appropriate immunoadsorbent to remove antibodies cross-reacting with human immunoglobulins. This renders it specific for use in test systems containing material of human origin (e. g. human tissue/mouse monoclonal antibody to an human tissue constituent/anti mouse Ig isotype specific immunoconjugate). Hyperimmune antisera with strong precipitating activity are selected for fractionation by salt-precipitation and purification of the IgG (7S) fraction by DEAE-chromatography.

Application Notes The cytochemical grade of GAM/mIg/7S allows its use in different types of highly sensitive immunoassays on appropriately treated cell and tissue substrates, in radioimmunoassay, for the production of immunoconjugates with a selected marker, to prepare immunoaffinity adsorbents by coupling to an artificial carrier, in non-isotopic methodology based on solid phase immunochemistry (e.g. ELISA), both as catching antibody and detection reagent, in Western blotting. This product is not pre-diluted.
The optimum working dilution of each product should be established by titration before being used. Working dilutions for histochemical and cytochemical use are usually between 1:100 and 1:250, in ELISA and comparable non-precipitating antibody-binding assays are between 1:500 and 1:5,000.

Reconstitution It is reconstituted by adding 1 mL sterile distilled water, spun down to remove insoluble particles, divided into small aliquots, frozen and stored at or below -24 °C.

Handling Advice Prior to use, an aliquot is thawed slowly at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7. 2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4 °C, not refrozen, and preferably used t he same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the product.
